how can i instruct Ad-Aware (Professional) to NOT scan a certain file (in the instant case, it's 12 GB)?

try to set it to ignore list.

To exclude a particular file, it would have to be in it's own folder. That way you could set up a Custom Scan, selecting all of the drives etc that you do wish to scan, but deselecting that particular 12 GB file.
Has to be in it's own folder, as the Custom Scan drop-down selection menu is not detailed enough to list every file in your system...
